Early morning sputum collection is particularly recommended for tuberculosis (TB) diagnosis due to its higher accuracy rate. This recommendation is based on the physiology of mucus production and how bacteria, including Mycobacterium tuberculosis, behave within the respiratory system.

During the night, the body produces mucus that can accumulate in the airways. When a patient first wakes up in the morning, the sputum collected is more likely to contain bacteria that have settled in the lungs overnight. This accumulation and the presence of more concentrated pathogens can enhance the sensitivity of tests conducted on the sample, leading to a more accurate diagnosis of TB when compared to sputum collected at other times of the day.
